II. Pharmacological properties

Proligestone is a steroid hormone that inhibits the secretion of gonadotropic hormones (especially luteinizing hormones), inhibits the maturation of follicles in the ovaries, blocks the release of estradiol from mature follicles and causes their regression, which prevents the development of estrus. Prolygestone causes changes in the uterine mucosa and increases the viscosity of mucus in the uterus, which prevents the implantation of a fertilized egg, inhibits cell hyperplasia and secretion of the mammary gland of animals. When administered during the anestrus phase, prolygestone maintains a low concentration of LH in the body, which prevents the growth of follicles and provides contraception for the animal. When the drug is discontinued, the concentration of prolygestone in the body decreases, restoring the normal level of LH in the animal’s blood, and the contraceptive effect of the drug ceases. Proligestone has very weak estrogenic, insignificant corticosteroid activity and does not have an androgenic effect.

9. The prolonged effect of the drug is associated with the deposition of prolygestone in fatty tissue and circulation in the liver. Prolygestone enters the intestine with bile, and most of it is removed with feces. A small amount of the hormone is excreted in the urine. The maximum concentration of prolygestone in plasma when administered subcutaneously reaches after 2-3.5 days and is 29 ng/ml. 13.1. To develop persistent anestrus in bitches and cats, Covinan is used in the indicated doses according to the basic scheme:

  • first injection during anestrus (for bitches and cats) or proestrus (for bitches);
  • second injection 3 months after the first injection;
  • third injection 4 months after the second injection.
  • then the drug is administered every 5 months.

If bitches who are treated with Covinan according to the specified regimen show signs of proestrus (pre-estrus), to prevent unwanted estrus, it is necessary to give an extraordinary injection of the drug in the appropriate dose.

The further regimen of use depends on the interval at which the injections were made before the appearance of undesirable signs of proestrus:

  • if the interval between regular injections was 5 months, then, after an extraordinary administration, Covinan is administered after 4 months, and then every 5 months:
  • if the interval was 4 months, then after an extraordinary injection, Covinan is subsequently administered according to the main regimen (after 3 months, then after 4 months, then the drug is administered every 5 months).

13.2. For one-time prevention of sexual heat in females and cats, Covinan is administered once to females during anestrus, preferably no later than a month before the expected estrus; for cats - Covinan is administered during the period of di- or anestrus.

13.3. To suppress estrus in bitches, Covinan is administered immediately after the appearance of signs of proestrus (bloody discharge and enlargement of the loop). Signs of proestrus gradually disappear within 5-7 days.

14. Restoration of the reproductive cycle in females occurs on average after 6-7 months; in rare cases, estrus is restored in the interval from 3 to 12 months.

15. When using the drug in rare cases (0.1-0.6% of cases), side effects such as pyometritis, endometrial hyperplasia, slight increase in body weight, increased appetite, apathy, and slight hypertrophy of the mammary glands are possible. The likelihood of side effects increases if other progesterone analogues were simultaneously administered to animals.
